FAQ: What is the difference between the NEBNext Ultra II Directional RNA Second Strand Module and the NEBNext mRNA Second Strand Synthesis Module?
The NEBNext Ultra II Directional RNA Second Strand Module includes the Second Strand Synthesis Reaction Buffer with dUTP Mix (dATP, dCTP, dGTP, and dUTP). The dUTP is used for labeling of the second strand for strand-specific library preparation by the Ultra Directional RNA method. The NEBNext mRNA Second Strand Synthesis Module contains Second Strand Synthesis Buffer that contains dATP, dCTP, dGTP and dTTP, and is not compatible with the Ultra Directional workflow.
